The Corynebacterium diphtheriae ISS 3319 strain was mutagenised with a mini-Tn5 transposon generating ~1 million mutants. The transposon-junctions of the C. diphtheriae mutants were sequenced to identify regions of the genome that can and cannot be disrupted, and therefore identify genes that are essential for viability.
