We report a strategy to personalize the use of radiotherapy on brain metastasis from lung cancer and breast cancer compatible with liquid biopsy and the use of a radiosensitizer. We perform 4 types of in vitro cultures using the lung adenoicarcinoma cell line H2030-BrM. These four culture conditions include regular adherent cultures (Adh), oncospheres (Onco), co-cultures between the cancer cell line and the glia using inserts (Insert) (the glia in the upper chamber; cancer cells in ower chamber) or mixed in the plate (Cocult). An additional condition included the administration of irradiation (10Gy) to all these four conditions. Irradiated and non-irradiated cultures (every culture in duplicate) were processed for RNAseq (Co-cultures plated in the same well underwent a sorting process based on the expression of GFP by cancer cells) 24 hours from irradiation.
