Gene expression profiling of purified stem and progenitor cell types from stabilized bicortical femoral fracture calluses one week after injury induction. We used microarrays to detail the global program of gene expression of stem and progenitor cells responsible for skeletal repair. Mouse skeletal stem and progenitor cells were purified from one-week femoral fracture calluses via flow cytometry for RNA extraction and hybridization on Affymetrix microarrays. We sought to obtain homogeneous populations of SSCs and BCSPs in order to compare the cell type specific expression profiles. To that end, we used 10-12 week-old mice and isolated CD45-Ter119-Tie2-CD51+CD200+6C3-Thy1-CD105- SSCs and CD45-Ter119-Tie2-CD51+6C3-Thy1-CD105+ BCSPs.
