Bulk RNA sequencing of Cd45+ cells from brains of aged Alzheimer's mice bearing human APOE knockin

Here we perform bulk RNA sequencing of immune cells of aged AD mice bearing knock-in of human APOE variants in the murine APOE locus. We produced single cell suspensions from the hippocampal and cortical regions of 60-week-old 5XFAD-hemizygous mice bearing homozygous knock-in of APOE2, APOE3, or APOE4 in the murine Apoe allele. Cells were sorted for Cd45+ directly into lysis buffer for RNA isolation. RNA was purified and used as input for SMARTseq low input library preparation.
