Nubp1_genome_sequencing

Mice harbouring a knockout allele for Nubpl (Nubpltm1b(KOMP)Wtsi) were produced by Wellcome Trust Sanger Institute Mouse Genetics Project (WTSI-MGP). Mice homozygous for this mutation die before weaning. Nubpltm1b(KOMP)Wtsi heterozygous mice were thus used for the pathogen challenge component of the WTSI-MGP. Upon challenge with S. typhimurium we found that mice from this background succumbed to infection by day 5-7 post infection presenting with a higher bacterial burden than WT control mice, which were on the same genetic background but from different knockout lines. Importantly we have subsequently determined that the mutation conferring susceptibility to S. typhimurium is not linked to Nubpl since wildtype littermates of Nubpltm1b(KOMP)Wtsi heterozygotes may also be susceptible. To find the susceptibility allele we aim to sequence the genomes of affected and non-affected mice.
