Two contagious cancer cell lines, representing Devil Facial Tumour 2 and Devil Facial Tumour 1 were cultured and DFT1 cells were treated with recombinant devil interferon gamma (IFNy). Cells were harvested and mRNA was extracted using the Nucleospin RNA mini kit (Machenery and Nagel) and following the manufacturers instructions. The quality of RNA was determined using the Agilent 2100 Bioanalyzer. 1000ng of RNA was used to generate first strand specific mRNA libraries (TruSeq Stranded mRNA kit from Illumina). Libraries were sequenced on an Illumina Hiseq by Eurofins Genomics, generating over 64 million 150bp paired end reads for each cell line. The RNAseq data generated was translated to protein and used the develop a custom protein database to identify peptides sequenced by mass spectrometry in the following study: Gastaldello A*, Ramarathinam SH*, Bailey A, Owen R, Turner S, Kontouli A, Elliott T, Skipp P, Purcell AW, Siddle HV. Transmissible cancers in the Tasmanian devil share a dominant peptide motif with the host. MS ID#: BIORXIV/2020/184416.
