α-Synuclein promotes H4R3me2s through interacting with BAF-PRMT5 complex

We conducted a screening for nuclear proteins which interact with α-synuclein by mass-spectrometry. As a result, we found that α-synuclein interact with BAF complex. Furthermore, overexpressed α-synuclein increased global H4R3me2s by promoting interaction between BAF complex and PRMT5 in cultered cells. In this study, we explored the influence of overexpressed α-synuclein on H4R3me2s by ChIP-seq using anti-H4R3me2s antibody. SH-SY5Y cells were neuronally differentiated over 10 days by culturing in 3%FCS DMEM supplemented wiith 10 µM RA for 6 days and DMEM without FCS supplemented with 50 ng/ml BDNF for following 4 days. α-synuclein was inducted for 3 days by 1 µg/ml doxycyline (Tet-on system). ChIP was conducted using H4R3me2s antibody.
