Proteome Profiler Human XL Oncology Array and Human Phospho-Kinase Array Kit

Under the condition of control and overexpression of PPM1B in HeLa cells, expression and phosphorylation level of numerous oncogenesis-related proteins were detected with R&D Systems Proteome Profiler Human XL Oncology Array kit and R&D Systems Proteome Profiler Human Phospho-Kinase Array kit (Minneapolis, MN, USA). Cells were treated, lysed as they are described in “PPM1B Protein Overexpression” and “Protein Extraction” sections. 200 μg of protein were applied followed the instructions of the manufacturer. Visualisation were performed by ChemiDoc Touch Imaging System. Dot intensities were quantified with ImageJ software. Raw values were corrected via subtraction of the background intensities. Final data were plotted as bars and heatmaps. Some significant hits were validated by semi-quantitative Western blot analysis or semi-quantitative RT-PCR.
