Cecal Microbiota of male WT and IL10 knockout fed control or High fat/high sucrose diets. Mus musculus

Male C57BL6/J mice that are either wildtype (WT) or knockouts (KO) for interleukin-10 gene were originally obtained from Jackson Laboratory. At four-weeks of age, both strains of mice (Wild and Knockout) were randomly assigned to one of two dietary treatments: WT + control diet (WT+CD, n=11), WT + High-fat diet (WT+WD, n=11), KO+CD (n=13) and KO+WD (n=12). (4 groups). Treatment was maintained for 12weeks, during which records of body weight and food intake were monitored. Mice were fasted for 3hours at the end of study, At the end of the study (30 days), mice were fasted for 3 hours, anaesthetized with ketamine/xylazine cocktail, bled from their carotid artery, and tissues were harvested. Each cecum was excised, flushed into individual tubes using ice cold sterile PBS, tubes were centrifuged, supernatant was discarded and the precipitate were weighed and stored at -80oC and prepped for 16S microbiota profiling.
