We have discovered that MEIS2 is aberrantly expressed in myeloma cells to predispose patients to adverse overall survival in IMiD therapy by suppressing the anti-myeloma interferon response and inhibiting the CRL4-CRBN E3 ubiquitin ligase in proximal IMiD signaling. Inhibition of CDK4/6 reverses both, uncovering a novel dual mechanism by which CDK4/6 inhibition augments IMiD's anti-myeloma activity and suggesting a potential clinical benefit to targeting CDK4/6 in combination with IMiD.
