Down-sampling analysis: Down-sampling analysis of network edge density between networks formed by different gene sets. P values were calculated by empirically estimating the probability of observing a denser network by randomly sampling 10,000 subnetworks of the same counts of genes from background interactome.CH-model: For a gene, we derive the expected number of de novo events in a given population based on the mutability of the gene and the number of probands sequenced, compare the observed number of de novo variants against expectation using a binormal model.
