To examine the difference of gene expression profiles between malignant melanoma cells treated with neopetasin (NP), metformin (Met), and phenformin (Phen), we have employed microarray expression profiling as a discovery platform. A2058 (human malignant melanoma) cells were treated for 8 hours with NP (3 µM), Met (5,00 µM), and Phen (50 µM). Gene expression in a human malignant melanoma cell line (A2058) treated with NP, Met, and Phen was measured at 8 hours after exposure to doses of 3, 5000, 50 µM of NP, Met, and Phen, respectively. Due to different solvent employed for each compound, profiles of the cells treated with NP was compared to that of DMSO; and, the profiles of the cells treated with Met and Phen were compared to that of water. Three independent experiments were performed.
