To understand the detailed dynamics of plants and microbes during infection, we deployed techniques to yield high-resolution RNA-seq using a 10x genomics single-cell workflow. Arabidopsis leaf tissue was inoculated by the fungal pathogen Colletotrichum higginsianum during biotrophic infection. plant cells were disassociated via an enzyme reaction and became protoplast as single cells. The single-cell suspension was loaded into 10x genomics chromium, and total mRNA from each loaded cell was barcoded and yielded after lysis reaction.
