The cardiac fibroblasts were isolated from heart of neonatal C57 mice. Cells at passages 2 to 3 were used to further experiments. Cells were incubated for 4-6 hours with serum-free DMEM. Transfection reagent iMax (Invitrogen, Carlsbad, CA, USA) was used for transfection of siRNA (siNC, siTmpo)and ASO (ASO-NC,ASO-Gm41724) (100 nM). 24 hours after transfection, cells were treated with TGF-beta-1 (10 ng/mL) for another 24 h.
