Impact of macrophage depletion and/or lupron (androgen deprivation therapy) on transcriptome of murine TMPRSS2-ERG+ expressing prostate tumors. Overall design: C57BL/6J mice were orthopically injected with 100,000 PTE-82 prostate cancer cells in 40 ul 1:1 PBS/Matrigel on Dec 2nd 2019. Indicated mice were randomized and treated with 0.6 mg Lupron / mouse on December 6th 2019. Indicated mice were treated with with a-CSF1 (anti-CSF1) on December 11th and December 16th 2019. On day 17 post-tumor injection, mice bearing orthotopic prostate tumors were cardiac perfused with PBS (without heparin), following by resection of tumors and immediate freezing in liquid nitrogen. RNA from prostate tumor tissue was extracted using RNeasy® Plus Mini kit (Qiagen) according to manufacturer's protocol with on-column DNase digestion and screened for quality on an Agilent Tape-Station (Agilent Technologies, Inc., Wilmington DE).
