We examined the application of RNA-Seq analysis to clarify the difference between the cell growth inhibitory effect of chemical and genetic knockdown on KRAS-mutant lung cancer cells. we identified that the treatment with verteporfin, a YAP1 inhibitor, reduced cell viability in KRAS-mutant lung cancer cells. To determine the role of YAP1 in these cascades, we carried out knockdown of YAP1 with siRNAs and assessed the effect on cell viability in KRAS-mutant lung cancer cells. Interestingly, the phenotypic effects of verteporfin and YAP1 knockdown seem that there is a difference, and RNA-seq results confirmed the difference from the genomic point of view. There was a difference in gene expression between treatment with verteporfin and knockdown with YAP siRNAs, and more regulators and pathways were involved especially in the cells treated with verteporfin. These analyses will provide additional insights into the pathogenesis of KRAS-mutant lung cancer cells.
