The MAIL8 cell line is a KBG-derived stable cell line that expresses ASC and a chimeric protein (C12N2) consisting of the CARD from CARD12 and the NOD and LRRs from Nod2 (NLRC2), a sensor for muramyl dipeptide (MDP). MDP stimulation induces C12N2-mediated oligomerization of ASC and cell-autonomous NF-kappa B and AP-1 activation, leading to IL-8 production in this cell line. The KBG cell line is a HEK293-derived stable cell line carrying a NF-kappa B-reporter gene expressing GFP. In this experiment, we stimulated MAIL8 cells with MDP to investigate the profile of the genes induced by ASC-activation. As controls, we investigated the gene expression profiles of KBG cells stimulated with MDP or TNF.
