The goal of this study is to investigate the effect of different sugar sources on microglial metabolism. Overall design: Murine N9 microglia were cultured in T75 flasks under control (standard physiological), high glucose (15 mM) and high fructose (15 mM) conditions for 18 hours. Microglia (250,000/well) were seeded in 24-well plates with the same media as in the T75 flasks and stimulated with LPS (1 µg/ml) for 6 hours. RNA was extracted and gene expression differences between the groups was determined using RNA-seq.
