SASI_seq

Sample assurance development This study involves development of SASI seq whereby tagged amplicons are spiked into samples destined for sequencing such that sample identity can be assured from the identity of those spike in tag sequences and any contamination or mix up can be spotted. A number of samples were sequenced on a number of different sequencing runs (mostly on Illumina MiSeq but two runs were done on HISeq). Runs were performed for the following tests Run 7701 Test of various size selection methodologies to see if SASI fragments could be lost Run 7790 Test of indexing oligos manufactured using Truegrade processing Run 7878 Sequencing of samples with SASI fragments following sureselect whole exome enrichment Run 7982 Test of various shearing protocols to see how SASI fragments were fragmented 8005 Test of indexing oligos manufactured using HPLC AND PAGE processing Run 8855 Sequencing run of one sample with barcode #2 Run 9357 Sequencing run of samples with barcode tags 190-193 Run 9699 Sequencing run of samples with odd numbered barcodes from 384 plex set Run 9728 Sequencing run of samples with even numbered barcodes from 384 plex set Run 10132, lane_5 Set of 96 multiplexed libraries each with unique SASI fragments Run 10132, lane_6 Set of 96 multiplexed libraries each with unique SASI fragments containing some mixup and cross contamination events
