MicroRNA-455 is a key microRNA in cartilage, strongly induced by Sox9. Overexpression of miR-455-3p in the developing chick limb bud at HH18-19 causes a reduction in limb bud size at HH24-25. Overall design: Limb buds were injected at H18-19 and harvested by dissection and snap freezing in liquid nitrogen after 24 hours at H24-25 prior to storage at -80oC Limb bud tissue was disrupted and homogenized in extractino buffer. RNA was purified using a combination Qiagen Rneasy Micro. We then performed RNASeq analysis on tissue from n=4 control mimic vs n=3 miR-455-3p mimic
