By utilizing Prss29-Cre, Pgr was conditionally deleted to identify the definitive roles of progesterone signaling in the uterine glands during pregnancy. This study provides new insights, demonstrating that PGR-P4 signaling in the glandular epithelium is required for uterine receptivity by regulating LIF on gestation day 4. Furthermore, we have shown that PGR signaling in glands is dispensable post-implantation. The uteri were collected from the wild-type (WT) and Prss29Cre/Cre;Pgrf/f (Pgr gecKO) on GD 4. RNA was extracted and sequenced by Azenta Life Sciences using two replicates for WT and Pgr gecKO. Libraries were prepared using the NEBNext Ultra II RNA Library Prep for Illumina and sequenced (2 x 150 bp paired end) using Illumina instrument (4000 or equivalent). Raw sequence data (.bcl files) generated from Illumina Hiseq was converted into fastq files and de-multiplexed using Illumina’s bcl2fastq 2.17 software.
