Notch signaling between neighboring cells vitally contributes to specifying their identity and function, during development and in the adult. Here, we demonstrate that selective blockade of the Notch ligands JAG1 and JAG2 disrupts thermoregulation in adults by decreasing circulating thyroid hormones and thyroid function. Sequencing data demonstrates that Notch inhibition decreases thyrocyte markers and mitochondrial genes. Overall design: Male mice of 10-12 weeks were treated with a single intraperitoneal injection of the Notch blocking antibodies (aJ1aJ2: 20mg/kg each; aN1aN2: 5, 10 mg/kg respectively; aN1aN2aN3: 5,10, 20 mg/kg respectively and anti-Ragweed isotype control: 40 mg/kg). Exogenous administration of thyroxine (T4) and 3-Iodotrionine (T3) was performed by intraperitoneal injection of 1ug/g BW of T4 and 0.05?g/g BW T3 diluted in PBS, daily starting at day 2 of the antibody treatment. Tissues were collected and snap frozen at the time of necropsy.
