Healthy human donor skin was dissociated, enriched for melanocytes using FACS and sequenced via smart-seq2 protocol. Donor age ranged from fetal stage up to later adulthood, which allowed for analysis of melanocyte development. In addition, various anatomical skin regions were sampled to explore site specificity of melanocyte transcription. The natural diversity of healthy human melanocytes was used to characterize bulk and single-cell melanoma samples to provide new insight into pathogenesis of the disease.
