The role of testicular macrophages (TMs), a diverse cell population crucial for tissue maintenance and immune balance, in fibrosis remains unclear. In this study, we utilized single-cell RNA sequencing (scRNA-seq) to investigate TM dynamics during UPEC infection in a murine model of epididymo-orchitis. Testes from UPEC-infected mice were collected on days 7 and 14, dissociated into single-cell suspensions, and CD45+ immune cells were sorted and encapsulated in oil droplets to form gel beads in emulsion using the DG1000 system.
