The OncoLoop Network-based Precision Oncology Framework

We introduce OncoLoop, a highly-generalizable, precision medicine framework to triangulate between available mouse models, human tumors, and large-scale drug perturbational assays with in vivo validation to predict personalized treatment Overall design: This series includes RNAseq from ten new and previously published genetically-engineered mouse models of prostate cancer. RNAseq from such primary tumors was used to construct a new interactome and to perform master regulator analysis that stratify individual tumors into molecular subtypes that are conserved with human prostate cancer tissues. By matching mouse models to patients and to drug reversion signatures, we validate that the OncoLoop algorithm predicts particular tumor vulnerabilities of individual tumors solely on transcriptome analysis.
