To establish a data-driven learning model of the temporal dynamics and 3D chromatin reorganization, we conducted tethered chromatin conformation (TCC) sequencing to examine 3D structure dynamics in estradiol (E2)-induced breast cancer T47D cells and Tamoxifen resistant breast cancer T47D cells. In this study, we conducted TCC analysis in hormone-starved T47D cells (T0) and compared this to T47D cells treated with 1hr of E2 (T1), 4hr of E2 (T4), 16hr of E2 (T16), 24hr of E2 (T24) and Tamoxifen resistant T47D cells. We could investigate dynamic 3D chromatin architecture in T47D cell lines from these TCC data.
