Chronic colitis has a significant negative impact on human health. The pathogenesis is unclear. The therapies for colitis are currently unsatisfactory. Probiotics have an anti-inflammatory property. This study aims to elucidate the role of Clostridium butyricum (CB)-derived DNA (DNAcb, in short) in attenuating colitis. We found that DNAcb induced colon epithelial cells to produce PDL1. DNAcb increased the quantity of KDM5A (a demethylase) in the PDL1 promoter to enhance the PDL1 gene transcription in colon epithelial cells. The colon epithelial cell-derived PDL1 induced activated Th1 cell apoptosis and attenuated experimental colitis. To sum up, DNAcb can induce colon epithelial cells to produce PDL1. The latter induces activated Th1 cell apoptosis and attenuates colitis
