Vicatia thibetica de Boiss polysaccharide 1 (VTP1) improves the myelosuppressive hematopoietic disorder and intestinal disorder induced by CTX in mice

Vicatia thibetica de Boiss polysaccharide 1 (VTP1) has the potential to improve myelosuppression induced by chemotherapy drugs. The aim of this study was to analyze the effects of VTP1 on myelosuppression through the combination of transcriptomics, gut microbiota and metabolomics.In vivo experimental results showed that VTP1 could improve blood cells caused by CTX, promote the recovery of spleen, thymus and femur, up-regulate the expression of hematopoietic factors (IL-3, G-CSF, EPO and TPO), promote the myeloid differentiation of hematopoietic stem cells, and maintain peripheral hematopoietic cells.Microbial sequencing results showed that VTP1 changed the proportion of microbial composition and increased the relative abundance of short-chain fatty acid production in Trichospirillaceae, Lactobacillaceae and Ruminococcaceae to improve intestinal disorders in myelosuppressed mice. Metabolomics analysis showed that VTP1 improved metabolic disorders in myelosuppressed mice by regulating bile acid secretion and amino acid synthesis.This study shows that VTP1 alleviates myelosuppression through different pathways, and the polysaccharide can affect characteristic microorganisms and metabolic pathways, but the specific mechanism of action needs further study.
