CBLL1, also known as HAKAI, plays a dual role in the METTL3-containing RNA m6A methyltransferase complex and as a regulator of e-cadherin protein expression. RNA methylation is known to play diverse functions in the regulation of mRNA processing, splicing, translation and stability. In this study we used siRNA to deplete CBLL1 in the MDA-MB-231 triple negative breast cancer cell line and the effect on global transcription determined. Overall design: A smartpool of four siRNAs (Dharmacon) were used to deplete CBLL1 in cultured MDA-MB-231 breast cancer cells and the effect on global transcription relative to a non-targetting siRNA scramble control determined using RNA sequencing.
