We developed a mouse model in which the pool of neural stem and progenitor cells (NSPCs) was partially ablated during early neurodevelopment by expressing attenuated diphtheria toxin A (DTA) in mCherry-negative NSPCs. This was achieved using a Cre-recombinase system controlled by the transgenic Nestin promoter (Nestin-Cre). In our model, the ablation is restricted to mCherry-negative NSPCs, allowing mCherry-positive NSPCs to continue their normal differentiation program. To assess the effects of this targeted reduction in NSPCs, we performed whole genome sequencing (WGS) at the end of cortical neurogenesis (E17.5), comparing both forebrain and tail regions.
