Proteomic analysis of vulnerable muscles (EDL) in the pre- and symptomatic stages of ALS

We used Tandem Mass Tags (TMT) 11-plex coupled to 2D-LC-MS/MS to profile EDL muscles from WT and SOD1G37R mice at presymptomatic (P380) and symptomatic (P480) stages. This strategy not only achieved high specificity and sensitivity, evidenced by robust enrichment of NMJ proteins and ALS-linked signatures, but also establishes a powerful framework to capture synaptic vulnerability and disease progression at the molecular level.
