We compared transcripts from whole brains dissected from stage 47 Xenopus laevis tadpoles from F0 NeuroD2 CRISPants and Cas9 injected control sibings. NeuroD2 knockdown brains had increased expression of cell cycle associated genes and decreased expression of chromatin modifing genes. Libraries were prepared and sequenced using Illumina Nextseq P2 by the Otago Genomics Facility, University of Otago, New Zealand. Single reads of 100bp were mapped to XENLA_10.1 genome build (Xenbase, accessed November 2024) Tadpoles were raised to stage 47 after injection at the one cell stage with either S. pyogenes Cas9 loaded with a frame shift inducing NeuroD2 sgRNA or just Cas9. Brains were removed by dissection and total RNA extracted from pooled groups of 4 using a qiagen RNeasy mini kit with on column DNase step. TruSeq stranded libraries were prepared. Single reads of 100bp Mapped to XENLA_10.1 genome build (Xenbase, accessed November 2024)
