Germ granules are RNA-rich biomolecular condensates containing factors for the posttranscriptional regulation of gene expression and the development of germ cells. In C. elegans, germ granules are associated with nuclear pore clusters. Previous studies have shown that assembly of P granules in the perinuclear region is critical for the fidelity of piRNA-mediated gene silencing. However, the mechanism by which germ granules associate with the nucleus and the functional relevance of such an association remain unknown. We uncovered the function of the NPP-14/24 complex in spatially organizing perinuclear germ granules and shed light on how organized germ granules function in fine-tuning the piRNA-based transcriptome surveillance and the inheritance of gene silencing.
