ATR inhibition synergizes with alkylating PI polyamide targeting MYCN by suppressing DNA repair in MYCN-amplified neuroblastoma

Because a single treatment with CCC-002 downregulated the expression of DDR-related genes in CHP-134 cells, the expression profile of SK-N-BE(2) cells after co-treatment with CCC-002 and an ATR inhibitor was evaluated using a microarray assay SK-N-BE(2) cells were treated with CCC-002 alone or in combination with AZD6738 for 24 h. RNA was extracted for microarray. Samples (150 ng) were labeled with an RNA Spike-In Kit and analyzed on SurePrint G3 Human GE 8 × 60 K V2 microarrays following the recommendations of Agilent Technologies. Sample arrays were scanned using an Agilent SureScan microarray scanner. The sample replicates were as follows: CCC-002, 3 × 2; DMSO, 3 × 2; and combination, 3 × 2.
