ATAC-seq of Vδ2+ γδ during controlled human malaria infection

γδ T cells play critical roles in innate immunity to Plasmodium falciparum malaria, yet their functional heterogeneity and memory-like dynamics during first and subsequent infections remain poorly defined. To investigate γδ T cell activation during malaria, we performed an Assay for Transposase-Accessible Chromatin sequencing (ATAC-seq) on Vδ2+ γδ T cells isolated from volunteers (n = 6) enrolled in a blood-stage P. falciparum CHMI trial. γδ T cells were sequenced at four timepoints: infection (I, 0 days post infection (d.p.i.)), anti-malarial drug treatment (T, 8 d.p.i.), and seven and nineteen days post treatment (15 d.p.i., 27 d.p.i.). Following quality control and peak analysis across timepoints, we generate a dataset of Differential Accessibility Regions (DARs).
