Testing heat-killed bacteria cluster formation by mixing two flouroscently labeled bacteria. Dye:Oregon green, excitation wave length: 488nm filter: FITC CF633-WGA, excitation wave length: 640nm filter Cy5 Each batch of single-stained bacteria has concentration of 100ul bacteria in 4ml PBS or serum mixture diluted 3% The concentration of the mixed batch would be the same as each single-stained batch (100ul in 4ml) file name type: mixture- type of buffer-constant shaking or stationary_time after mixing Example: mixture-PBS-CS-4h Codes: constant shaking: CS Stationary: S type of imaging : flouroscent: FS Parameters of experiments: biological state: Heat-killed solution resting state: constant shaking, stationary Time of leaving the sample for cluster formation: Intervals of 4h is considered. Type of solution: pooled serum 2.5 v/v% diluted by PBS or serum magnification: 20X Pixel size : 16um camera and microscope setting sola power 9% for FITC and 20% for Cy5 exposure: 1 frame PFs (autofocus is on) optical configuration was one with elham_fitc and elham_cy5 scan 3*3 and 3 times in ND aquisition not in aquire and fast tiune-lapse [in calibration tab}
