We established a micro-patterned respiratory epithelial cell culture system in vitro. In this culture system, the iPSC-derived lung progenitor cells were differentiated into airway epithelial cells in a position-specific manner. The human iPSC-derived lung progenitor cells were seeded onto micro-patterned plates and cultured for 7 days. Nuclei were stained with Hoechst-33342, and the Edge and Center cell populations of the colony were separated and collected according to the Hoechst intensity. The Hoechst-high intensity population corresponded to the peripheral cells of the colony (Edge cells), and the Hoechst-low intensity population corresponded to the central cells of the colony (Center cells).
