The aim of this study was to explore the role of quercetin and isoquercetin in hepatic gluconeogenesis, using the human liver tumor cell line HepG2. The mRNA expression of PEPCK, G6Pase, and LKB1 was evaluated using RT-qPCR. The protein expression of LKB1, TORC2, P-TORC2, AMPK, p-AMPK, SIK1, and p-SIK1 was evaluated using western blotting.
本研究以人类肝肿瘤细胞系HepG2(human liver tumor cell line HepG2)为实验模型,旨在探究槲皮素(quercetin)与异槲皮素(isoquercetin)在肝脏糖异生(hepatic gluconeogenesis)中的调控作用。通过实时荧光定量聚合酶链式反应(RT-qPCR)检测了磷酸烯醇式丙酮酸羧激酶(PEPCK)、葡萄糖-6-磷酸酶(G6Pase)及肝激酶B1(LKB1)的mRNA表达水平;采用蛋白质免疫印迹(western blotting)分析了LKB1、TORC2、P-TORC2、腺苷酸活化蛋白激酶(AMPK)、磷酸化腺苷酸活化蛋白激酶(p-AMPK)、盐诱导激酶1(SIK1)及磷酸化盐诱导激酶1(p-SIK1)的蛋白表达水平。
