Chromatin fragments from HeLa cell extract by sonication were incubated with the DMC1-ssDNA complex or the RAD51-ssDNA complex. The chromatin fragments associated with the DMC1-ssDNA complex or the RAD51-ssDNA complex were pelleted using the magnetic beads conjugated at the edge of the ssDNA. The genomic DNA fragments associated to the DMC1-ssDNA complex or the RAD51-ssDNA complex were then analyzed by deep sequencing.
