CFSE-labeled PBMCs were incubated with TT and MBP for 7 days, restimulated with PMA/ ionomycin and brefeldin for 5 hours, permeabilized and stained for surface markers and intracellular cytokines with fluorochrome-conjugated antibodies. The percentage of interleukin (IL)-17A, IFN-γ, IL-4, tumor necrosis factor (TNF)-α, IL-13 and IL-10 expressing cells within CFSE-diluted (proliferating) CD3+CD8- T-cell population was assessed by flow cytometry.
