Dendritic cells (DCs) simultaneously prime and dampen the immune response. The transcriptional program that regulates immunosuppression in DCs is currently unknown. Here we identified ETS homologous factor as a key transcription factor that regulates DC immunosuppression. Overall design: Mice were immunized intraperitoneally (i.p.) with or without 60 µg TLR stimulation in 100 µL PBS. Splenic cells were collected 18 hrs after injection for cDCs (live MHCII+ CD11c+) FACS-sorting for scRNA-seq library preparation.
