Generation of Syngeneic Orthotopic DMG Murine Models

Diffuse Midline Glioma (DMG) syngeneic orthotopic murine mouse models were used as high fidelity model of human DMG tumors as they recapitulate the heterogeneity, cell states and OncoTarget/OncoTreat drug predictions of human DMG tumors. Overall design: Single-cells were isolated and sequenced from B6(Cg)-Tyrc-2J/J (B6) mice. An intracranial injection with 100,000 DIPG4423 cells was performed and tumor was isolated 3 weeks post-injection for single cell. Additionally, single cells were also isolated from normal brainstem (no tumor) of the same model
