Partial LFQ Proteomic Dataset from Myocardial Infarcted Heart Tissues Following Transplantation of Pre-differentiated and Undifferentiated Umbilical Cord Tissue-Derived Mesenchymal Stem Cells
收藏资源简介:
This study was conducted in accordance with ARRIVE guidelines and approved by the institutional bioethical committee, University of Karachi, Pakistan. All animal experiments were performed in accordance with institutional guidelines for the care and use of laboratory animals. Human umbilical cord samples were collected from healthy donors who underwent elective cesarean after taken their consent agreement. Human umbilical cord derived Mesenchymal stem cells (UC-MSCs) were isolated and cultured under standard culture conditions. A subset of UC-MSCs was transiently transfected with GATA-4 and MEF2C expression plasmids at 37 °C with 5% CO2 for 48 hours and then cultured for 14 days to induce cardiac pre-differentiation. Myocardial infarction (MI) model was developed by ligation of the left anterior descending (LAD) coronary artery. Animals were randomly assigned into four experimental groups (n = 3 per group): Sham control Myocardial infarction (MI) MI + GATA-4 and MEF2C guided pre-differentiated UC-MSCs transplantation MI + un-differentiated UC-MSCs transplantation After four weeks of pre-differentiated and un-differentiated cell transplantation, infarcted and peri-infarct regions of the left ventricle were harvested for quantitative proteomic analysis.
