Small-Angle X-ray Scattering (SAXS) Data of OmpLA Reconstituted in POPC and DLPC Proteoliposomes

This dataset contains Small-Angle X-ray Scattering (SAXS) profiles for Large Unilamellar Proteoliposomes (PLUVs) containing Outer Membrane Phospholipase A (OmpLA). The liposomes were prepared via detergent removal by dialysis, using lipid systems of either POPC + 5 mol% POPG or DLPC + 5 mol% DLPG. Data Acquisition Two distinct beamline/instrument setups were utilized depending on the lipid system: POPC-based samples: Measured at the BioSAXS beamline P12 (EMBL Hamburg, PETRA III, DESY, Germany). Samples (~7 mg/mL) were flowed through a 1 mm quartz capillary at 37°C. Data reduction was performed on-site using standard EMBL pipelines. Targeted protein/lipid molar ratios included 1/900, 1/600, and 1/300. DLPC-based samples: Measured using a SAXSpoint 5.0 system (Anton Paar, Graz, Austria) equipped with a MetalJet X-ray generator and an Eiger R 1 M detector. Samples (~20 mg/mL) were held in an Anton Paar μCell at 37°C. Data integration and background subtraction were performed using SAXSanalysis software (Anton Paar). Targeted ratios included 1/1200 and 1/300. Reference systems: Mock control data (lipid-only vesicles processed identically to protein samples). Buffer composition: 20 mM TRIS buffer + 2 mM EDTA. Nomenclature PL: protein to lipid molar ratio (e.g. DLPC_OMPLA_PL_300: OmpLA in DLPC vesicles at a molar ratio of 1/300) POPC: 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine POPG: 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphoglycerol DLPC: 1,2-dilauroyl-sn-glycero-3-phosphocholine DLPG: 1,2-dilauroyl-sn-glycero-3-phosphoglycerol Context Full methodological details regarding PLUV preparation and concentration determination can be found in the associated manuscript supplementary materials.
