Detection of RNA bound by RIG-I in 1205Lu cells infected by yellow fever (YF)17D vaccine virus by in vivo proximity labeling

1205Lu cells expressing human RIG-I fused to a monomeric streptavidin tag were infected with 10 MOI YF17D for 48 hrs post. Cells were incubated with Biotin-TFPA-PEG3 tag or left untagged and UV-irradiated for photolabeling. RNA was isolated and incubated with streptavidin beads. RIG-I/RNA complexes were recovered by streptavidin pull-down. cDNA libraries were generated after removal of rRNA and sequenced using Illumina NextSeq 500 platform.
