In this project, we engineer a contractile injection system (CIS) from Photorhabdus asymbiotica to load custom payload proteins and deliver them with high specificity into defined target cells, including human cell lines, primary cells, and live mice. Our results demonstrate that CISs are programmable protein delivery devices with possible applications in gene therapy, cancer therapy, and biocontrol.
