We built a high-throughput optimization platform for intracellularly active antimicrobial peptides that allows rapid investigation of a large part of the antimicrobial fitness landscape. Using deep mutational scanning, we experimentally explored the sequence space of Bac7 without the need for excessive chemical synthesis and developed a highly potent bacterial ribosomal inhibitor.
