In this study, we aimed to determine the effects of combination therapy on drug-resistance emergence in herpes simplex virus 1 (HSV-1). The wild-type HSV-1 KOS strain was grown under increasing doses of a single antiviral (acyclovir, ganciclovir, trifluridine) or combinations thereof (acyclovir/trifluridine and ganciclovir/trifluridine). The viral thymidine kinase and DNA polymerase were genetically characterized by amplicon-based next-generation sequencing on the Illumina Miseq platform.
