We measured bulk mRNA abundance in Ngn2 induced patient derived cortical neurons in synucleinophaty models. ActD shuts off the transcription allowing us to measure mRNA decay. Overall design: Biological samples: SNCA 2 copy neurons and SNCA 4 copy neurons, Treatment: ActD(10 ug/ml), Collection times: 0,1,3,6,12 hour after ActD tretment, Perturbation: SNCA preformed fibril addition(5 ug/ml)
