Genomic analysis of multidrug resistant Klebsiella pneumoniae isolated from patients in Chattogram, Bangladesh

The emergence of multidrug resistant Klebsiella pneumoniae (KPN) is a global concern due to increasing rate of mortality and hospital cost burden in the affected population. The present study aimed to characterize the genome of multidrug-resistant KPN isolates from clinical specimens by whole genome sequencing (WGS). The isolates of KPN (n = 10) were collected from clinical samples of hospitals located at Chattogram region in Bangladesh. The bacterial isolates were cultured, and antimicrobial susceptibility testing was performed following CLSI guidelines. The genomes of all MDR isolates were analyzed by retrieving WGS technique data on the using Illumina iSeq100 platform. Generated reads were trimmed and subjected to assembly generation using SPAdes software. The assembled contigs were further analysed in Kleborate for identifying sequence type, antibiotic resistance genes, virulence factors and capsule type (K type) and serotype (O type) in KPN genomes. This is the first attempt to conduct a genomic study on the pathogenic bacteria in the Chattogram of Bangladesh. The molecular characterization of isolated KPN provided substantial evidence of existing antimicrobial resistance (AMR) in the bacterial species. The WGS annotations for the isolates might be utilized for evidence-based antibiotic stewardship in the region.
